Just to stop watching tv I replayed an old favorite game series of mine, the Crysis line.
In Crysis warhead the Australian character you play, affectionately known as "Psycho", tells a sadistic murderous north korean colonel "I'm British, you muppet." before leaving him to the tender mercies of a hostile alien vessel the size of a city block.
In the original Deus Ex (still a great game) J.C.Denton tells the main enemy who just bragged that after seizing power he would burn like the brightest star "You're gonna burn, all right."
I don't play a huge number of video games, so if you know any lines from them you think are good let's share them. I'm sure there were some good ones in bioshock 1 and 2 but don't recall them now.

One of the first fully voiced games I played - Bioforge - pretty much opened with:
"We should not be enemies!" beats man with his own severed arm "We should work together!" beats man with own arm "I have no wish to harm you!" kicks man in face
25 years on and the silliness of it still sticks with me.
I don't play more than a couple of videogames a year, including those ones that I already finished it (some even 10+ times).
Above everything I put the Mass Effect trilogy.
But I'm also still doing the old Baldur's Gate and Vampires and once every 3-4 years I have a complete run of those titles, so I'm very excited to try the upcoming games from these sagas.
The old Blizzard games had some really funny ones. Like the elf wizard in warcraft 3 going "for ultimate spell of destruction press control, alt, delete".

Also when numerous charcters in a bunch of computer games:

Followed one mission later with the very matter-of-fact "The prisoner did not survive the interrogation."
As a comedy line I'd have to go with "Can I have some shoes?" from the GLA worker in CnC generals.
As a serious line I loved "And from that throne, I... will watch over you" from Arcturus Mengsk's inauguration speech in Starcraft. It was just the culmination of such a brilliant character arc.
"So far are we along the path... I struggle to hear our enemies as they crowd around us" from the Prophet of Truth in Halo 3 and "Such is the fate, Dun Dun, of the enemies of Man" at the end of DoW2 campaign missions also get special mentions!

Almost all of the dialogue in Banjo-Kazooie is pretty funny and quotable.
Referring back to Halo 2 like others have said, I'd have to go with Sgt. Johnson's line, "For a brick, he flew pretty good!"

For pure feels... Mordin Solus. "Had to be me. Someone else might have gotten it wrong." Specifically, the last time he says it in Mass Effect 3.
But my absolute favourite has to be Joker in Arkham City - who, for context, is slowly dying from blood poisoning. Not the voicemails (which are absolute gold) but his cutscene line:
"Take my blood, for example. I wish somebody would. This stuff is killing me!"

There is a lack of most of Portal and its sequel in this thread. Cave, lemons, mantis men, companion cubes. Too many to list.
Psychonaughts also has some incredible lines, particularly in the Milkman Conspiracy. The cookies gag is up there, but never forget, "it's fortified with what the world needs.... with what the world... DESERVES"
